PURPOSE: A novel brain targeting drug delivery system based on OX26 antibody conjugation on PEGylated cationic solid lipid nanoparticles (OX26-PEG-CSLN) was prepared. METHODS: The Baicalin-loaded PEGylated cationic solid lipid nanoparticles modified by OX26 antibody (OX26-PEG-CSLN) were prepared by emulsion evaporation-solidification at low temperature method. The immune-gold labeled OX26-PEG-CSLN was visualized by transmission electron microscopy. The mean diameter and zeta potential of OX26-PEG-CSLN, PEG-CSLN and CSLN were determined using a Zetasizer. The entrapment efficiency of OX26-PEG-CSLN, PEG-CSLN and CSLN was determined by ultrafiltration centrifugation method. And the solid-state characterization of OX26-PEG-CSLN and CSLN were analyzed by X-ray. Pharmacokinetics studies were conducted by in vivo microdialysis in rat cerebrospinal fluid. RESULTS: The results showed that the OX26-PEG-CSLN, PEG-CSLN and CSLN had average diameters of 47.68 ± 1.65, 27.20 ± 1.70 and 33.89 ± 5.74 nm, Zeta potentials of -0.533 ± 0.115 mV, 11.200 ± 0.500 mV and 11.080 ± 1.170 mV and entrapment efficiencies of 83.03 ± 0.01%, 92.90 ± 3.50% and 97.83 ± 0.19%, respectively. In the pharmacokinetics studies, the AUC value of OX26-PEG-CSLN was11.08-fold higher than that of the Baicalin solution (SOL) (p<0.01), and 1.12-fold higher than that of the CSLN (p>0.05); the Cmax value of OX26-PEG-CSLN was 7.88-fold higher than that of SOL (p<0.01) and 1.12-fold (p<0.01) higher than that of the CSLN, respectively. CONCLUSION: These results demonstrated OX26-PEG-CSLN could be a promising carrier to deliver drugs across the BBB for the treatment of brain diseases.
STUDY OBJECTIVES: To evaluate the efficacy and safety of acupuncture at HT 7 (Shenmen) and KI 7 (Fuliu) on sleep and comorbid symptoms for chronic insomnia. METHODS AND DESIGN: A randomized, single-blind, parallel and sham-controlled trial consisted of an acupuncture group (n = 41) and a sham acupuncture group (n = 41). Setting: a tertiary hospital of integrated Chinese and Western medicine. Participants: 82 subjects with chronic insomnia based on the International Classification of Sleep Disorders, Third Edition (ICSD-3). Interventions: a 10-session acupuncture treatment at bilateral HT 7 and KI 7 or sham acupoints with shallow needling was performed over 3 weeks. Measurements: the Pittsburgh sleep quality index (PSQI) and insomnia severity index (ISI) were evaluated at baseline, posttreatment, and at two follow-ups as the primary outcome measures. Polysomnography (PSG) on two consecutive nights, the Beck anxiety inventory (BAI), the Beck depression inventory (BDI) fatigue severity scale (FSS) and the Epworth sleepiness scale (ESS) were evaluated at baseline and posttreatment as the secondary outcome measures. RESULTS: After the treatments, PSQI scores decreased by 5.04 in the acupuncture group and 2.92 in the sham acupuncture group. ISI scores decreased by 7.65 in the acupuncture group and 5.05 in the sham acupuncture group. The between-group differences in the primary outcome measures posttreatment were statistically significant. However, no differences were found between the two groups during the two follow-ups. Regarding the PSG data, there were significantly lower levels of sleep onset latency (SOL), a lower percentage of sleep stage N1 and a higher percentage of sleep stage N3 in the acupuncture group than in the sham acupuncture group. After treatment, there were lower levels of comorbid symptoms (BAI, BDI, FSS and ESS) in both groups. However, no significant differences were observed between the groups. CONCLUSION: Acupuncture at HT 7 and KI 7 is an effective and safe nonpharmacologic intervention option for chronic insomnia. The current study aimed to investigate the effects and mechanisms of electroacupuncture (EA) treatment applied to Bai hui (GV20) and Yin tang (GV29) acupoints (1 mA, 2 Hz, continuous wave, 20 minutes) for 28 days in a rat model of chronic unpredictable mild stress (CUMS) on reuptake of serotonin (5-hydroxytryptamine (5-HT)) and miRNA-16 levels in the hippocampus and serum. Rats were housed in individual cages, and CUMS was used to establish a rat model of depression. After EA treatment for 4 weeks, behavioral changes and indices including 5-HT transporter (SERT), 5-HT, and miRNA-16 levels in the hippocampus and serum were examined. The EA treatment significantly improved base levels of sucrose preference and exploratory behavior and significantly decreased SERT protein and mRNA expression in the hippocampus of depressed rats. Significantly increased 5-HT levels were observed, and miRNA-16 levels were significantly decreased in the hippocampus and serum of depressed rats. In conclusion, the antidepressant effects of EA treatment may be affected via inhibition of 5-HT reuptake, upregulation of 5-HT levels, and inhibition of miRNA-16 expression in the hippocampus and serum.
BACKGROUND: Chronic simple pharyngitis (CSP) is a common clinical chronic respiratory inflammation with persistent and intransigent symptoms. We analyzed the clinical data to find the evidence that herbal tea, a traditional Chinese medicine treatment in China, could improve the symptoms of CSP patients in a simple way. METHODS: We systematically reviewed the clinical data of randomized controlled treatments from April 2019 and evaluated the results using the improved Jadad scale and the Cochrane bias risk assessment tool. RevMan 5.3 software was used for chart analysis. In addition, we used Excel to conduct frequency statistics on Chinese herbs from included articles and analyze its medication rules. RESULTS: Among the collection of 161 articles, 6 RCTs published in Chinese journals were included in this review. The methodological quality of the treatments was low, and most of them only provide diagnostic criteria. Inclusion and exclusion criteria were not specified, and none of the 6 RCTs used the blind method on the result evaluator. Furthermore, only one RCT evaluated the baseline level variance. For these reasons, we did not make a network meta-analysis. CONCLUSIONS: The traditional Chinese herbs involved in herbal tea did have ingredients to alleviate CSP symptoms. However, our research showed that the current research could not draw any credible conclusions on the curative effect of herbal tea, which indicated that the overall level of TCM clinical research needs to be improved to evaluate the efficacy of herbal tea.
BACKGROUND: Insomnia is often related to stressful events. The hypothalamus-pituitary-adrenal (HPA) axis is related to stress, and dopamine (DA) and DA receptors are involved in the regulation of HPA axis. Electroacupuncture (EA) can improve sleep in individuals with insomnia, but the mechanism is unclear. We demonstrated that EA can improve sleep in rats after cage change through DA and the DA receptors in the HPA axis. METHODS: A rat model of insomnia was established by cage change to a dirty cage. The rats in treatment groups were intervened by EA and D1R (or D2R) antagonists. Electroencephalography (EEG) and electromyogram (EMG) were recorded to compare the changes in sleep. The DA, corticotropin-releasing hormone (CRH), adrenocorticotropic hormone (ACTH), and cortisol (CORT) levels in the plasma and hypothalamus were measured by ELISAs, and the D1R and D2R levels were measured by RT-PCR and immunohistochemistry. RESULTS: The dirty group showed a significant increase in the amount of wakefulness and decrease in the amount of NREM sleep, with decreased numbers of long NREM sleep bouts and REM sleep bouts and increased mean duration of wakefulness during the light period. EA and D1R (or D2R) antagonists intervention could improve sleep disturbance by decreasing wakefulness in the light period after cage change, EA and D1R (or D2R) antagonists could increase the hypothalamus DA, CRH, ACTH, CORT level, and the D1R and D2R mRNA levels in the HPA axis, and the effect of EA plus D1R (or D2R) antagonist was not superior to that of EA or D1R (or D2R) antagonists alone. CONCLUSIONS: EA can improve the sleep of rats after cage change, and the mechanism may be related to the regulation of DA and D1R or D2R in the HPA axis.
INTRODUCTION: Insomnia with short sleep duration has a more serious negative impact on patient health. The existing literature suggests that medication therapy is more effective for this phenotype of insomnia compared with cognitive-behavioural therapy. However, the potential side effects of hypnotic medications hinder their clinical application. Acupuncture has been widely used in the treatment of insomnia, but it remains unclear whether it has therapeutic efficacy for insomnia with short sleep duration. The purpose of this trial is to evaluate the efficacy and safety of acupuncture for insomnia with short sleep duration. METHODS AND ANALYSIS: This study is designed as a randomised, single-centre, single-blinded, placebo acupuncture controlled trial involving 152 participants. Eligible patients will be divided into two groups according to the objective total sleep time: insomnia with normal sleep duration group and insomnia with short sleep duration group. Then, patients in each group will be randomly assigned to two subgroups, the treatment group (acupuncture) and the control group (placebo acupuncture), in a 1:1 ratio with 38 subjects in each subgroup. The primary outcome is the Pittsburgh Sleep Quality Index and the Insomnia Severity Index. Secondary outcomes are actigraphy, the Beck Anxiety Inventory, the Beck Depression Inventory and the Fatigue Severity Scale. All adverse effects will be assessed by the Treatment Emergent Symptom Scale. Outcomes will be evaluated at baseline, post treatment, as well as at 1-week and 1-month follow-up. ETHICS AND DISSEMINATION: This protocol has been approved by the ethics committee of Yueyang Hospital of Integrated Traditional Chinese and Western Medicine (no. 2019-17). Written informed consent will be obtained from all participants. The results will be disseminated through peer-reviewed journals for publications. Coumestrol facilitates apoptosis in CRC cells by interacting with ZIP8 protein via the ferroptosis pathway.
Doxorubicin (DOX) is an anthracycline class of chemotherapy drug, the application of which is limited due to its cardiotoxic effects. Recombinant Proprotein Convertase Subtilisin/Kexin Type 9 (PCSK9), is a serine protease pivotal in lipid metabolism and has a profound correlation with the onset of cardiovascular diseases. This study uncovers a connection between PCSK9 and DOX-induced cardiotoxicity (DIC). This research found that injection of DOX in mice caused cardiac toxicity. DOX treatment up-regulated the expression of PCSK9 protein in myocardial tissue. Evolocumab (PCSK9 inhibitors) improved cardiac function, myocardial injury, and fibrosis in DOX-treated mice, indicating a protective effect against DIC. The mechanism involved modulation of cardiomyocyte apoptosis and regulation of apoptosis-related proteins, including Bax/Bcl-2 ratio and Cleaved Caspase-3/Pro Caspase-3 ratio. DOX exhibited concentration- and time-dependent cytotoxic effects on H9C2 cardiomyocytes, promoting apoptosis. PCSK9 nuclear aggregation occurred in H9C2 cardiomyocytes after DOX treatment, and PCSK9 interacted with the Importin subunit beta-1 (KPNB1) protein. Interference with PCSK9 up-regulated KPNB1 expression, affecting apoptosis-related proteins and improving DOX-induced H9C2 cardiomyocyte apoptosis. In short, the elucidation of this mechanism is helpful involve that PCSK9 inhibitor may be a potential drug for improving DIC.

Abstract Cobalt-doped Fe3O4 was prepared by a simple oxidation–precipitation method. Characterization results confirm that Co was successfully doped into Fe3O4. The doped Co increased the surface area, surface hydroxy groups and positive charge density of the product, which accelerated the decomposition of ozone into hydroxyl radicals. Cobalt-doped Fe3O4 exhibited high catalytic activity for the mineralization of 2,4-D by ozone in aqueous solution.
Background Hypertension is the major global preventable risk factor in cardiovascular diseases. While traffic noise is a highly disruptive environmental stressor in contemporary cities, its longitudinal causal association with hypertension remains debatable in past research. This study aims to conduct a systematic review and meta-analysis of high-quality cohort studies to evaluate the correlation between long-term traffic noise exposure and the risk of hypertension in adults. Methods PubMed, Embase, and Web of Science databases were systematically searched to identify relevant published literature. Prospective or retrospective cohort studies examining the relationship between long term exposure to traffic noise with the occurrence of hypertension in adults were incorporated. The quality of the research studies was evaluated based on the Newcastle-Ottawa Scale methodologically. The association was estimated by data pooling with fixed-or random-effect models and estimated by calculating the HR and its 95%CI. Results 11 cohort studies with 1,353,481 participants were included into the present research. The results indicated that in cohorts analyzing traffic noise as a binary variable, higher exposure was significantly associated with an increased risk of hypertension (HR = 1.07, 95% CI: 1.01–1.13, P = 0.03). This finding is highly consistent with the meta-analysis results where traffic noise was treated as a continuous variable (HR = 1.03, 95% CI: 1.02–1.04, P < 0.00001). While heterogeneity was low in the main analysis, moderate to high heterogeneity was present in specific subgroups, such as night noise, follow-up time, and aircraft noise. Subgroup analyses revealed that the pathogenic effect of traffic noise was robust across different spatial and temporal noise sources, as well as follow-up time, and completely independent of traditional confounding factors such as air pollution. Conclusion Long-term exposure to traffic noise is significantly associated with an increased risk of hypertension in the adult population. Incorporating environmental noise assessment and mitigation measures into public health strategies can contribute to the comprehensive management of hypertension in the population. BaiYangJie (BYJ) is a terrestrial perennial plant commonly used as a Dai medicine and has therapeutic effects on liver and kidney diseases. Cisplatin (CP), a chemotherapy drug, has good therapeutic effects but causes many side effects, including nephrotoxicity. This article investigated the toxicology of the methanol extract of BYJ (ME-BYJ) and its protective effect on CP-induced acute kidney injury (AKI) through pharmacological experiments. The results showed that the treated mice had no toxicological symptoms and no anatomical, physiological, or histological abnormalities. The BYJ-high-dose group showed significantly attenuated CP-induced AKI. It is concluded that ME-BYJ has the most significant protective effect on AKI at a dose of 8 g/kg and BYJ was not toxic.
The rapid development of information technology has provided more method choices for the reform of college English teaching.At the same time, due to the impact of the Novel Coronavirus in the spring semester of 2020, nationwide online live teaching has greatly promoted the form of "Internet + education".Based on the teaching practice of the author and other teachers in the same college, this paper analyses in details the common problems in the current college English live classes, proposes some suggestions for improvement, and finally discusses how to build college English "online and offline" blended teaching model based on online live courses in the future.
Objective To investigate of compassion fatigue and occupational stress among ICU nurses in Class Ⅲ Grade A hospitals, and to explore the correlation. Methods From April to May 2017, a total of 106 ICU nurses from 2 Class Ⅲ Grade A hospitals in Zhangjiakou were recruited in the research. All the participants were investigated with a General Condition Questionnaire, the Chinese version of the Compassion Fatigue Scale and the Occupational Stress Sources of Nurses Questionnaire. SPSS 22.0 software was applied in data analysis, and Pearson correlation analysis methods was used to explain the correlation between compassion fatigue and occupational stress. Results The total scores of compassion fatigue and occupational stress were (61.57±21.44) and (93.19±17.49) respectively in ICU nurses. There were significant differences in compassion fatigue scores among nurses of different age, working years and professional titles (t/F=3.718, 6.893, 2.818; P<0.05) ; there were significant differences in occupational stress scores among nurses of different working years (F=3.364, P<0.05) . Compassion fatigue was positively correlated with occupational stress (P<0.01) . Conclusions The compassion fatigue in ICU nurses is at a high level. The compassion fatigue is positively correlated with occupational stress. It is suggested that nursing managers should pay attention to the occupational stress and compassion fatigue among ICU nurses, and take necessary interventions to reduce them, so as to improve the nursing quality effectively. Houttuynia cordata Thunb. is a medicinal and edible herb. It can be used as a snack and also treat many diseases such as pneumonia, influenza, tumors, and so on. It contains volatile oil, flavonoids, alkaloids, polysaccharides, and other active ingredients. In this study, its active ingredients and clinical applications were introduced through comparative analysis and summary of extensive literature. Conclusion: Houttuynia cordata Thunb. has extremely high research potential due to its efficacy in treating various diseases with minimal side effects.
Urban infrastructure networks underpin modern supply chain operations, yet their vulnerability to disruptions poses significant challenges to urban resilience; understanding and predicting such disruptions requires analytical frameworks that jointly leverage network topology and flow dynamics. This study proposes a three-stage framework integrating complex network analysis, Graph Convolutional Network (GCN)-based predictive modeling, and resilience stress testing, validated on a large-scale New York City dataset comprising 6,302 infrastructure nodes, 96,641 disruption events, and 100,000 traffic flow records. The proposed GCN model achieves an RMSE of 5.8 and R2 of 0.92, significantly outperforming ARIMA, SVR, LSTM, and XGBoost baselines, with ablation experiments confirming that graph structure reduces prediction error by 29.3%. Resilience analysis reveals that targeted hub-node attacks cause rapid initial fragmentation, while a crossover effect at approximately 30% node removal shows that random failures induce greater cumulative damage. Temporal factors and historical flow patterns are identified as the dominant predictors, collectively accounting for 65.5% of feature importance. The integrated framework bridges flow prediction and resilience assessment, enabling identification of dual-risk nodes for prioritized infrastructure protection and providing actionable insights for enhancing urban supply chain resilience.
BACKGROUND: Kinesiophobia may reduce participation in post-stroke rehabilitation and impede functional recovery, but its course after discharge and its clinical significance remain unclear. This study identified 6-month trajectories of kinesiophobia in patients with post-stroke hemiplegia and examined their associations with motor recovery and rehabilitation exercise adherence. METHODS: In this single-center prospective longitudinal study, 296 patients with post-stroke hemiplegia were assessed at discharge and at 1, 3, and 6 months after discharge. Baseline discharge assessment occurred a mean of 18.7 ± 5.8 days after stroke onset, so the follow-up window captured both early subacute and later community recovery phases. Kinesiophobia was measured with the validated Chinese Tampa Scale for Kinesiophobia (TSK-17), motor recovery with the Fugl-Meyer Assessment (FMA), and rehabilitation exercise adherence with the validated Chinese Engagement in Exercise Rehabilitation Scale for Patients with Stroke (EERS), assessed from month 1 onward. Self-report measures could be completed by standardized face-to-face or telephone interview when outpatient attendance was not possible, whereas FMA was assessed only by trained raters during outpatient or home-based in-person visits. Kinesiophobia trajectories were identified using latent class growth analysis. Associations with repeated FMA and EERS outcomes were analyzed using adjusted linear mixed-effects models, and baseline predictors of trajectory membership were examined with a bias-adjusted three-step multinomial logistic model. RESULTS: <.001). At 6 months, adjusted mean FMA scores were 74.2, 65.9, and 50.6 across the three classes, and corresponding EERS scores were 70.5, 66.8, and 55.8. Compared with the persistent-low class, the persistent-high class had markedly poorer motor recovery (mean difference, 23.6 points; 95% CI, 19.2-28.0) and lower exercise adherence (mean difference, 14.7 points; 95% CI, 10.8-18.6), while the decreasing-moderate class showed intermediate deficits. Older age, history of falls, higher anxiety and depression, and lower exercise self-efficacy independently predicted unfavorable trajectories. CONCLUSIONS: Post-discharge kinesiophobia follows heterogeneous trajectories in patients with post-stroke hemiplegia. Persistently high kinesiophobia identifies a vulnerable subgroup with poorer motor recovery and lower rehabilitation exercise adherence. Early screening and targeted support may improve post-discharge rehabilitation outcomes.
Enzyme-Linked Immunosorbent Assay (ELISA) has become one of the most widely used immunological detection technologies and plays an important role in the detection of human diseases. This paper reviews the research background, core position, types, basic principle, producer and the progress of the ELISA, and based on the principle and the procedure to research the clinical application of the ELISA in human diseases focusing on the tumor screening and detection, infectious diseases and allergy. Also analyse the strengths, weaknesses and the future prospect of the ELISA. The result of the research demonstrate that although there some limitation with sometimes have the rate of missed diagnosis and false diagnosis and some cannot be done simultaneously, it is a better way to develop some technology like the high-throughput ELISA technology, and we can also make the combination with ELISA and nucleic acid testing method. With high sensitivity, strong specificity, low cost and easy standardization, ELISA can serve as a powerful technical support for clinical detection and diagnosis of human diseases.
Background: Chronic hepatitis B (CHB) remains a major cause of cirrhosis and hepatocellular carcinoma. Xiaochaihu decoction (XCHD) is frequently used in China as an adjunct to Western medicine (WM), but its incremental benefit remains uncertain. This review evaluated the efficacy and safety of XCHD added to WM in patients with CHB. Methods: Eight databases were searched from inception to October 5, 2025, for randomized controlled trials comparing XCHD + WM with the same background WM alone. Full search strategies are provided in Supplementary Material A. Primary outcomes were virological response (HBV-DNA, HBeAg, and HBsAg negativity) and liver biochemical indices (ALT and AST). Secondary outcomes included total clinical efficacy, TBil, HA, LN, PCIII, IV-C, and adverse events. Risk of bias was assessed with RoB 2, certainty of evidence was assessed with GRADE, and trial sequential analysis (TSA) was used as a supplementary assessment of random error. Results: Forty-nine trials (n = 4,768) were included. Compared with WM alone, XCHD + WM was associated with higher HBV-DNA, HBeAg, and HBsAg negativity rates and lower ALT, AST, TBil, HA, LN, PCIII, and IV-C values. Total clinical efficacy was higher, and the reported incidence of adverse events was not increased. However, most continuous outcomes showed substantial heterogeneity, several outcomes were affected by suspected publication bias, and certainty was low or very low for most biochemical and fibrosis-related outcomes. Discussion: Current evidence suggests that XCHD may offer short-term adjunctive benefits for surrogate virological and biochemical outcomes when added to WM for CHB, without an apparent increase in reported adverse events. While these preliminary findings are promising, due to methodological limitations and heterogeneity, future large-scale and high-quality RCTs are warranted to confirm their long-term clinical benefits and establish definitive evidence.
This dataset supports the manuscript "Predictive analytics for supply chain resilience in urban infrastructure networks using graph convolutional networks" (submitted to PLOS ONE, 2026). The study proposes a three-stage analytical framework integrating complex network analysis, Graph Convolutional Network (GCN)-based flow prediction, and network resilience stress testing, validated on a New York City urban road network dataset. The GCN model achieves an RMSE of 5.8 and R² of 0.92 for supply chain flow prediction, outperforming ARIMA, SVR, LSTM, and XGBoost baselines. Resilience analysis identifies a crossover effect at approximately 30% node removal where random failures surpass targeted attacks in cumulative connectivity loss.
