Effects of COVID-19 Home Confinement on Eating Behaviour and Physical Activity: Results of the ECLB-COVID19 International Online Survey

BACKGROUND: Public health recommendations and governmental measures during the COVID-19 pandemic have resulted in numerous restrictions on daily living including social distancing, isolation and home confinement. While these measures are imperative to abate the spreading of COVID-19, the impact of these restrictions on health behaviours and lifestyles at home is undefined. Therefore, an international online survey was launched in April 2020, in seven languages, to elucidate the behavioural and lifestyle consequences of COVID-19 restrictions. This report presents the results from the first thousand responders on physical activity (PA) and nutrition behaviours. METHODS: Following a structured review of the literature, the "Effects of home Confinement on multiple Lifestyle Behaviours during the COVID-19 outbreak (ECLB-COVID19)" Electronic survey was designed by a steering group of multidisciplinary scientists and academics. The survey was uploaded and shared on the Google online survey platform. Thirty-five research organisations from Europe, North-Africa, Western Asia and the Americas promoted the survey in English, German, French, Arabic, Spanish, Portuguese and Slovenian languages. Questions were presented in a differential format, with questions related to responses "before" and "during" confinement conditions. RESULTS: 1047 replies (54% women) from Asia (36%), Africa (40%), Europe (21%) and other (3%) were included in the analysis. The COVID-19 home confinement had a negative effect on all PA intensity levels (vigorous, moderate, walking and overall). Additionally, daily sitting time increased from 5 to 8 h per day. Food consumption and meal patterns (the type of food, eating out of control, snacks between meals, number of main meals) were more unhealthy during confinement, with only alcohol binge drinking decreasing significantly. CONCLUSION: While isolation is a necessary measure to protect public health, results indicate that it alters physical activity and eating behaviours in a health compromising direction. A more detailed analysis of survey data will allow for a segregation of these responses in different age groups, countries and other subgroups, which will help develop interventions to mitigate the negative lifestyle behaviours that have manifested during the COVID-19 confinement.

O presente estudo busca reunir informações acerca da Base Nacional Comum Curricular, demonstrando que essa Base define as competências e conhecimentos essenciais para orientar a formação do aluno de ensino médio. O presente artigo tem por finalidade, expor analise com referencia a capacidade da BNCC de orientar os alunos na escolha das áreas de formação e dos itinerários formativos, após um levantamento de pesquisas por meio do Google Acadêmico e varias leituras em relação a BNCC. O artigo expõe as analises feitas com base nas propostas da BNCC, de opiniões e criticas de profissionais da educação, no sentido de esmiuçar os meandros da nova fase da educação no Brasil, que passará por transformações no anos vindouros, seja na parte de metodologia do ensino, seja na área do desenvolvimento pedagógico. Tema que no momento traz preocupações, sobre a autonomia das redes de ensino em construir e disponibilizar seus próprios currí­culos. O estudo concluiu que É importantí­ssimo que os alunos compreendam cada competência, é importante também conversar sobre cada uma delas, discutir com os alunos o que eles estão aprendendo, e porque estão aprendendo, para que entendam o sentido, a importância de cada área de formação e que possam estar abertos a colaborar para que juntos os estudantes possam ajudar a desenvolver o coletivo, sendo protagonistas de suas historias. Se torna muito importante o diálogo e as discussões sobre os projetos polí­ticos pedagógicos das escolas com todos os professores, gestores e colaboradores, para que a concepção de educação possa ser compreendida e incorporada, tornar-se salutar, aproveitar este momento para reflexão sobre a formação do ser humano que é o aluno nesta fase da sua vida.

Effects of home confinement on mental health and lifestyle behaviours during the COVID-19 outbreak: Insight from the ECLB-COVID19 multicenter study

Although recognised as effective measures to curb the spread of the COVID-19 outbreak, social distancing and self-isolation have been suggested to generate a burden throughout the population. To provide scientific data to help identify risk factors for the psychosocial strain during the COVID-19 outbreak, an international cross-disciplinary online survey was circulated in April 2020. This report outlines the mental, emotional and behavioural consequences of COVID-19 home confinement. The ECLB-COVID19 electronic survey was designed by a steering group of multidisciplinary scientists, following a structured review of the literature. The survey was uploaded and shared on the Google online survey platform and was promoted by thirty-five research organizations from Europe, North Africa, Western Asia and the Americas. Questions were presented in a differential format with questions related to responses "before" and "during" the confinement period. 1047 replies (54% women) from Western Asia (36%), North Africa (40%), Europe (21%) and other continents (3%) were analysed. The COVID-19 home confinement evoked a negative effect on mental wellbeing and emotional status (P < 0.001; 0.43 ≤ d ≤ 0.65) with a greater proportion of individuals experiencing psychosocial and emotional disorders (+10% to +16.5%). These psychosocial tolls were associated with unhealthy lifestyle behaviours with a greater proportion of individuals experiencing (i) physical (+15.2%) and social (+71.2%) inactivity, (ii) poor sleep quality (+12.8%), (iii) unhealthy diet behaviours (+10%), and (iv) unemployment (6%). Conversely, participants demonstrated a greater use (+15%) of technology during the confinement period. These findings elucidate the risk of psychosocial strain during the COVID-19 home confinement period and provide a clear remit for the urgent implementation of technology-based intervention to foster an Active and Healthy Confinement Lifestyle AHCL).

Psychological consequences of COVID-19 home confinement: The ECLB-COVID19 multicenter study

All participants were asked for their mental wellbeing (SWEMWS) and depressive symptoms (SMFQ) with regard to "during" and "before" home confinement. RESULTS: Analysis was conducted on the first 1047 replies (54% women) from Asia (36%), Africa (40%), Europe (21%) and other (3%). The COVID-19 home confinement had a negative effect on both mental-wellbeing and on mood and feelings. Specifically, a significant decrease (p < .001 and Δ% = 9.4%) in total score of the SWEMWS questionnaire was noted. More individuals (+12.89%) reported a low mental wellbeing "during" compared to "before" home confinement. Furthermore, results from the mood and feelings questionnaire showed a significant increase by 44.9% (p < .001) in SMFQ total score with more people (+10%) showing depressive symptoms "during" compared to "before" home confinement. CONCLUSION: The ECLB-COVID19 survey revealed an increased psychosocial strain triggered by the home confinement. To mitigate this high risk of mental disorders and to foster an Active and Healthy Confinement Lifestyle (AHCL), a crisis-oriented interdisciplinary intervention is urgently needed.

Accountability: já podemos traduzi-la para o português?

O objetivo principal deste artigo é verificar se as alterações políticas, sociais e institucionais (ocorridas no Brasil) contribuíram para que a tradução da palavra accountability germinasse no solo brasileiro, tendo transcorrido duas décadas desde a publicação do instigante artigo de Anna Maria Campos sobre a ausência desse conceito no Brasil. Trata-se de estudo de natureza exploratória, analítica e descritiva, numa abordagem essencialmente qualitativa, em que se procurou, além de compreender o significado da palavra accountability nos dicionários e nos trabalhos sucessivos ao de Campos, analisar, por meio da literatura especializada, as principais mudanças processadas no cenário brasileiro, especialmente quanto à organização da sociedade, descentralização e transparência governamental e quanto à emergência de novos valores sociais em substituição aos tradicionais. Reconhecendo que avanços têm sido realizados nessa direção, admite-se ser difícil dar uma resposta conclusiva à questão formulada. Considera-se que estamos mais perto da resposta do que quando Campos se defrontou com o problema, mas ainda muito longe de construir uma verdadeira cultura de accountability.

The efficacy of clove oil as an anaesthetic and in euthanasia procedure for small-sized tropical fishes

Clove oil is used as a fish anesthetic because it is a natural and inexpensive product with low toxicity risks. The goal of the present study was to determine the appropriate concentration of clove oil for small-sized tropical fish to be used in mark-recapture studies or when individuals are to be sacrificed. We applied three different clove oil concentrations (D1=0.05 mL, D2=0.10 mL and D3=0.20 mL per 500 mL of water) on three small-sized fish species. We found a negative relationship between induction time and treatment for two species (Hyphessobrycon sp.1 and Hemigrammus sp.), while concentration was unrelated to recovery time. Fish body length was positively related to induction time in the D2 treatment for Hemigrammus sp., and negatively for Hyphessobrycon sp.1 in the D1 treatment, but was unrelated to recovery time for three species and treatments. Mortality rates varied across treatments, but higher rates were observed with higher clove oil concentrations. We conclude that 0.05 mL of clove oil per 500 mL of water is the most efficient dose for studies where fish will be released back to their natural habitats, while 0.20 mL of clove oil is recommended for studies that require fish euthanization for further laboratory analyses.

Globally altered sleep patterns and physical activity levels by confinement in 5056 individuals: ECLB COVID-19 international online survey

Symptoms of psychological distress and disorder have been widely reported in people under quarantine during the COVID-19 pandemic; in addition to severe disruption of peoples' daily activity and sleep patterns. This study investigates the association between physical-activity levels and sleep patterns in quarantined individuals. An international Google online survey was launched in April 6 th , 2020 for 12-weeks. Forty-one research organizations from Europe, North-Africa, Western-Asia, and the Americas promoted the survey through their networks to the general society, which was made available in 14 languages. The survey was presented in a differential format with questions related to responses "before" and "during" the confinement period. Participants responded to the Pittsburgh Sleep Quality Index (PSQI) questionnaire and the short form of the International Physical Activity Questionnaire. 5056 replies (59.4% female), from Europe (46.4%), Western-Asia (25.4%), America (14.8%) and North-Africa (13.3%) were analysed. The COVID-19 home confinement led to impaired sleep quality, as evidenced by the increase in the global PSQI score (4.37 2.71 before home confinement vs. 5.32 3.23 during home confinement) (p < 0.001). The frequency of individuals experiencing a good sleep decreased from 61% (n = 3063) before home confinement to 48% (n = 2405) during home confinement with highly active individuals experienced better sleep quality (p < 0.001) in both conditions. Time spent engaged in all physical-activity and the metabolic equivalent of task in each physical-activity category (i.e., vigorous, moderate, walking) decreased significantly during COVID-19 home confinement (p < 0.001). The number of hours of daily-sitting increased by ~2 hours/days during home confinement (p < 0.001). COVID-19 home confinement resulted in significantly negative alterations in sleep patterns and physical-activity levels. To maintain health during home confinement, physical-activity promotion and sleep hygiene education and support are strongly warranted.

Condições associadas ao grau de satisfação com a vida entre a população de idosos

OBJECTIVE: As a result of overall growing population's life expectancy, it has become increasingly important to ensure not only that the elderly have greater longevity but also happiness and life satisfaction. The objective of the study was to describe factors associated with life satisfaction among elderly people. METHODS: Three hundred and sixty-five older persons, selected by means of random stratified proportional sampling, were interviewed in 2003. The instrument used was a combination of Flanagan and Nahas questionnaires and WHOQOL-100. There were added questions concerning physical activity extracted from International Physical Activity Questionnaire, questions regarding reported morbidity and emotional assessment, sociodemographic condition and an open question. The level of life satisfaction was measured using a scale from one to seven by means of visual recognition. Hierarchical logistic regression analysis was performed including "life satisfaction" as a dependent variable and those included the final questionnaire, in blocks, as independent variables. RESULTS: Most elderly were generally rather satisfied with life as well as with specific aspects. The level of life satisfaction was associated with: comfort at home (OR=11.82; 95% CI: 3.27; 42.63); appraising leisure as quality of life (OR=3.82; 95% CI: 2.28; 6.39); waking up feeling well in the morning (OR=2.80; 95% CI: 1.47; 5.36); not reporting loneliness (OR=2.68; 95% CI: 1.54; 4.65); having three or more daily meals (OR=2.63; 95% CI: 1.75; 5.90) and not reporting Diabetes Mellitus (OR=2.63; 95% CI: 1.31; 5.27). CONCLUSIONS: Most elderly in the study were satisfied with life and their satisfaction was associated with situations related to "being well" and not being diabetic.

Biofilm formation is the preferred mode of growth lifestyle for many microorganisms, including bacterial and fungal human pathogens. Biofilm is a strong and dynamic structure that confers a broad range of advantages to its members, such as adhesion/cohesion capabilities, mechanical properties, nutritional sources, metabolite exchange platform, cellular communication, protection and resistance to drugs (e.g., antimicrobials, antiseptics, and disinfectants), environmental stresses (e.g., dehydration and ultraviolet light), host immune attacks (e.g., antibodies, complement system, antimicrobial peptides, and phagocytes), and shear forces. Microbial biofilms cause problems in the hospital environment, generating high healthcare costs and prolonged patient stay, which can result in further secondary microbial infections and various health complications. Consequently, both public and private investments must be made to ensure better patient management, as well as to find novel therapeutic strategies to circumvent the resistance and resilience profiles arising from biofilm-associated microbial infections. In this work, we present a general overview of microbial biofilm formation and its relevance within the biomedical context.

Reporting Guidelines for Whole-Body Vibration Studies in Humans, Animals and Cell Cultures: A Consensus Statement from an International Group of Experts

Whole-body vibration (WBV) is an exercise modality or treatment/prophylaxis method in which subjects (humans, animals, or cells) are exposed to mechanical vibrations through a vibrating platform or device. The vibrations are defined by their direction, frequency, magnitude, duration, and the number of daily bouts. Subjects can be exposed while performing exercises, hold postures, sitting, or lying down. Worldwide, WBV has attracted significant attention, and the number of studies is rising. To interpret, compare, and aggregate studies, the correct, complete, and consistent reporting of WBV-specific data (WBV parameters) is critical. Specific reporting guidelines aid in accomplishing this goal. There was a need to expand existing guidelines because of continuous developments in the field of WBV research, including but not limited to new outcome measures regarding brain function and cognition, modified designs of WBV platforms and attachments (e.g., mounting a chair on a platform), and comparisons of animal and cell culture studies with human studies. Based on Delphi studies among experts and using EQUATOR recommendations, we have developed extended reporting guidelines with checklists for human and animal/cell culture research, including information on devices, vibrations, administration, general protocol, and subjects. In addition, we provide explanations and examples of how to report. These new reporting guidelines are specific to WBV variables and do not target research designs in general. Researchers are encouraged to use the new WBV guidelines in addition to general design-specific guidelines.

Prevalência de Sintomas de Ansiedade e Depressão em Estudantes de Medicina

Objetivos Determinar a prevalência de sintomas de ansiedade e depressão em estudantes de Medicina e avaliar fatores associados. Métodos Estudo de corte transversal com 234 estudantes que responderam a um questionário eletrônico com variáveis sociodemográficas e a Escala Hospitalar de Ansiedade e Depressão (Ehad). Resultados Em relação à ansiedade, o escore médio da Ehad foi de 6,7 (DP: +/- 3,4), com 34,3% (80) apresentando sintomas falso-positivos de ansiedade e 19,7% (46) manifestando sintomas sugestivos do transtorno. Quanto à depressão, o escore médio da Ehad foi de 4,4 (DP: +/- 3,1), com 19,3% (45) apresentando sintomas falso-positivos para depressão e 5,6% (13) manifestando sintomas sugestivos do transtorno. Na análise univariada, o uso de drogas psicoativas associou-se à presença de sintomas de ansiedade; para sintomas de depressão, o estudante ser procedente da RMR foi fator protetor, ao passo que o uso de drogas ilícitas foi associado a risco. Conclusão A prevalência de sintomas de ansiedade e depressão associada ao uso de drogas psicoativas e ilícitas, respectivamente, indica a necessidade de medidas de prevenção e diagnóstico precoces.

Epithelial Mesenchymal Transition Induces Aberrant Glycosylation through Hexosamine Biosynthetic Pathway Activation

Deregulated cellular metabolism is a hallmark of tumors. Cancer cells increase glucose and glutamine flux to provide energy needs and macromolecular synthesis demands. Several studies have been focused on the importance of glycolysis and pentose phosphate pathway. However, a neglected but very important branch of glucose metabolism is the hexosamine biosynthesis pathway (HBP). The HBP is a branch of the glucose metabolic pathway that consumes ∼2-5% of the total glucose, generating UDP-GlcNAc as the end product. UDP-GlcNAc is the donor substrate used in multiple glycosylation reactions. Thus, HBP links the altered metabolism with aberrant glycosylation providing a mechanism for cancer cells to sense and respond to microenvironment changes. Here, we investigate the changes of glucose metabolism during epithelial mesenchymal transition (EMT) and the role of O-GlcNAcylation in this process. We show that A549 cells increase glucose uptake during EMT, but instead of increasing the glycolysis and pentose phosphate pathway, the glucose is shunted through the HBP. The activation of HBP induces an aberrant cell surface glycosylation and O-GlcNAcylation. The cell surface glycans display an increase of sialylation α2-6, poly-LacNAc, and fucosylation, all known epitopes found in different tumor models. In addition, modulation of O-GlcNAc levels was demonstrated to be important during the EMT process. Taken together, our results indicate that EMT is an applicable model to study metabolic and glycophenotype changes during carcinogenesis, suggesting that cell glycosylation senses metabolic changes and modulates cell plasticity.

Prevalence of comorbidities in patients and mortality cases affected by SARS-CoV2: a systematic review and meta-analysis

The new coronavirus, COVID-19 was declared a pandemic by the World Health Organization on March 11, 2020. Risk factors associated with this disease are age, sex, and the presence of comorbidities, the most common being hypertension, diabetes, and heart disease. The aim of this meta-analysis was to calculate the prevalence and geographical distribution of comorbidities in all patients admitted to intensive care units (ICUs), and the mortality rate of COVID-19. We selected studies based upon epidemiological and clinical descriptions of the patients and mortality from the disease to determine the pooled prevalence of comorbidities in all patients and in mortality cases due to COVID-19. The pooled prevalence was estimated using the random effects model, and odds ratios were used to measure the probability of death for a patient with a comorbidity. The total prevalence of comorbidities in patients with COVID-19 was 42% (95% CI: 25-60), 61% (95% CI: 42-80) in those admitted to the ICU, and 77% (95% CI: 68-86) among death cases; males were the most affected. Hypertension was the most prevalent comorbidity in all three groups studied, accounting for 32%, 26%, and 35%, respectively. The odds ratio of death for a patient with a comorbidity compared to one with no comorbidity was 2.4 (P < 0.0001). The higher the prevalence of comorbidities the higher the odds that the COVID-19 patient will need intensive care or will die, especially if the pre-existing disease is hypertension, heart disease, or diabetes.

p53 reactivation with induction of massive apoptosis-1 (PRIMA-1) inhibits amyloid aggregation of mutant p53 in cancer cells

p53 mutants can form amyloid-like structures that accumulate in cells. p53 reactivation with induction of massive apoptosis-1 (PRIMA-1) and its primary active metabolite, 2-methylene-3-quinuclidinone (MQ), can restore unfolded p53 mutants to a native conformation that induces apoptosis and activates several p53 target genes. However, whether PRIMA-1 can clear p53 aggregates is unclear. In this study, we investigated whether PRIMA-1 can restore aggregated mutant p53 to a native form. We observed that the p53 mutant protein is more sensitive to both PRIMA-1 and MQ aggregation inhibition than WT p53. The results of anti-amyloid oligomer antibody assays revealed that PRIMA-1 reverses mutant p53 aggregate accumulation in cancer cells. Size-exclusion chromatography of the lysates from mutant p53-containing breast cancer and ovarian cell lines confirmed that PRIMA-1 substantially decreases p53 aggregates. We also show that MDA-MB-231 cell lysates can "seed" aggregation of the central core domain of recombinant WT p53, corroborating the prion-like behavior of mutant p53. We also noted that this aggregation effect was inhibited by MQ and PRIMA-1. This study provides the first demonstration that PRIMA-1 can rescue amyloid-state p53 mutants, a strategy that could be further explored as a cancer treatment.

Prevalência de depressão em idosos que freqüentam centros de convivência

The objective of the study was to establish the prevalence rate of depression among the elderly population (those 60 and older) who frequent community centers. From February to July of 2001, a cross-sectional survey was conducted with elderly people from community centers in Taguatinga, Brasilia, FD (Federal District). The sample included 118 elderly people, distributed in five-year age brackets, who responded to a simplified version of the Yesavage Geriatric Depression Scale with 15 questions. Anova and the Tukey test were performed to analyze differences between the age groups with 95% confidence intervals. The sample was predominantly female (90%) with the majority reporting being in the age bracket of 60 to 64 years old (31%). Depression was reported by 36 respondents (31%) and severe depression was reported by 4% of those interviewed, all of whom were in the age bracket of 60 to 64 (representing 14% of this group). National programs should be developed in community centers focused on decreasing depression among the elderly population.

Using a Game Engine for VR Simulations in Evacuation Planning

A game engine is applied to virtual simulations of emergency evacuation from a modeled building of Brazil's Nuclear Engineering Institute (IEN). Researchers tested evacuation scenarios to compare the virtual simulations to real simulations using real people in the real building. The results compared favorably and provide a basis for further experiments to improve user interfaces and avatar autonomy.

Qualidade de vida em estudantes de Medicina no início e final do curso: avaliação pelo Whoqol-bref

O objetivo do estudo é comparar a qualidade de vida de estudantes de Medicina do primeiro período e do último período do curso. Foi realizado um estudo transversal com a aplicação do instrumento da Organização Mundial da Saúde para avaliação da qualidade de vida no seu formato curto (Whoqol-bref) em 370 estudantes de Medicina da cidade do Recife, Brasil - 229 do primeiro e 141 do último período. Os resultados mostraram que os escores do domínio psicológico foram menores nos alunos do último período do que nos do primeiro período (p < 0,005). Essa diferença se manteve após a análise pelo modelo de covariância (Ancova). Em relação aos escores do domínio físico, relações sociais e meio ambiente, não houve diferenças significativas entre os alunos do primeiro e do último período. Quanto à autoavaliação da qualidade de vida, os alunos do primeiro período apresentaram melhores resultados (3,87 vs. 3,39; p < 0,001). Quanto à satisfação com a própria saúde, não houve diferença estatística; 3,94 vs. 3,62 (p = 0,099). Em conclusão, a qualidade de vida dos estudantes de Medicina, quando avaliada pelo instrumento Whoqol-bref, sofre desgastes no domínio psicológico durante o curso médico. Novos estudos são necessários para identificar os fatores que determinam essas alterações na qualidade de vida dos estudantes de Medicina durante a graduação.

Fatores associados ao óbito hospitalar por COVID-19 no Espírito Santo, 2020

OBJECTIVE: To analyze factors associated with the deaths of individuals hospitalized with COVID-19 in the state of Espírito Santo, Brazil. METHODS: This was a cross-sectional study using secondary data. Logistic regression models were used to estimate crude and adjusted odds ratios (OR). RESULTS: As at May 14, 2020, 200 individuals had been discharged and 220 had died. Of the total number of people studied, 57.1% were male, 46.4% were >60 years old, 57.9% were cases notified by a private institution, and 61.7% had >1 comorbidity. In the adjusted analysis, hospital mortality was higher for the 51-60 age group (OR=4.33 - 95%CI 1.50;12.46), the over 60 age group (OR=11.84 - 95%CI 4.31;32.54), cases notified by public institutions (OR=8.23 - 95%CI 4.84;13.99) and cases with a greater number of comorbidities (two [OR=2.74 - 95%CI 1.40;5.34] and three [OR=2.90 - 95%CI 1.07;7.81]). CONCLUSION: Higher mortality was found in older individuals, those with comorbidities and users of public hospitals.

Effects of Selenium Supplementation in Patients with Mild Cognitive Impairment or Alzheimer’s Disease: A Systematic Review and Meta-Analysis

Elevated levels of oxidative stress could cause and aggravate Alzheimer's disease (AD). Selenium (Se) is a trace element with antioxidant and anti-inflammatory activity with neuroprotective effects. To evaluate the effects of Se supplementation in patients with AD or mild cognitive impairment (MCI) through a systematic review and meta-analysis, data were searched and collected from four electronic databases, including clinical trial studies published until December 2020, following the PRISMA guidelines. Statistical analysis was performed by RevMan, and the risk of bias was assessed using the Rob 2 tool. A total of 1350 scientific papers were collected, and following evaluation 11 papers were included in the systematic review and 6 of these were used in the meta-analysis. Studies that evaluated only Se supplementation observed an improvement in Se levels, glutathione peroxidase (GPX) activity, and in some cognitive tests in MCI patients; similarly, improvement in Se levels and mini-mental score was also observed in AD patients. Regarding supplementation of Se plus other nutrients, improvement in cognitive tests was observed in both AD and MCI patients. Therefore, Se supplementation is a good alternative for patients with AD and MCI for improving Se levels and GPX activity. More detailed studies are required to further evaluate the effects of Se on the cognitive deficit and oxidative stress associated with AD and MCI.
