This textbook, edited by Jane Ritchie and Jane Lewis, is meant for both students and researchers, but because it primarily presents basic knowledge it is more suitablefor students. It is intended to lead practitioners throughthe process of qualitative research, i.e. from the design of astudy, conducting of in-depth interviews and analysis of datato the presentation of results. The authors impart in a professionalway both broad theoretical knowledge and practice-oriented information. They do not provide the readerwith an overview of qualitative methods, but focus on indepthinterviews and so-called focus groups.
BACKGROUND/AIMS: Several recent studies revealed an accumulation of ceramide in bronchial, tracheal and intestinal epithelial cells of mice and patients with cystic fibrosis (CF). Normalization of ceramide concentrations in lungs of CF mice employing the functional acid sphingomyelinase inhibitor amitriptyline also normalized mucociliary clearance, chronic inflammation and infection susceptibility to pulmonary P. aeruginosa in these mice. METHODS: To test for a beneficial effect of amitriptyline in vivo, we performed a phase IIb randomised, double-blind, placebo-controlled study. Twenty-one CF patients were treated with 25 mg/d amitriptyline twice daily for 28 days. The placebo consisted of 19 patients and was also treated twice per day. The primary endpoint was the change in lung function in the intention-to-treat (ITT) population. Secondary endpoints were ceramide levels in epithelial cells and safety. RESULTS: After treatment, forced expiratory volume in 1 sec predicted (FEV1) increased 6.3 ± 11.5% (p=0.08) in the ITT population (36 of 40 CF patients) and 8.5 ± 10% (p=0.013) in the per protocol (PP) population (29 of 40 patients). Ceramide levels decreased in nasal epithelial cells after amitriptyline treatment. Amitriptyline had no severe and only mild and mostly transient adverse effects, i.e. xerostomia and tiredness. CONCLUSION: Amitriptyline is safe in CF-patients, increases FEV1 and reduces ceramide in lung cells of CF patients.
The Large Hadron Collider (LHC) at CERN will extend the frontiers of particle physics with its \nunprecedented high energy and luminosity. Inside the LHC, bunches of up to 1011 protons (p) \nwill collide 40 million times per second to provide 14 TeV proton-proton collisions at a design \nluminosity of 1034 cm2s1. The LHC will also collide heavy ions (A), in particular lead nuclei, at \n5.5 TeV per nucleon pair, at a design luminosity of 1027 cm2s1. \nThe high interaction rates, radiation doses, particle multiplicities and energies, as well as the \nrequirements for precision measurements have set new standards for the design of particle detectors. \nTwo general purpose detectors, ATLAS (A Toroidal LHC ApparatuS) and CMS (Compact \nMuon Solenoid) have been built for probing p-p and A-A collisions. \nThis paper presents a comprehensive overview of the ATLAS detector prior to the first LHC \ncollisions, written as the installation of the ATLAS detector is nearing completion. This detector \nrepresents the work of a large collaboration of several thousand physicists, engineers, technicians, \nand students over a period of fifteen years of dedicated design, development, fabrication, and installation.
Precipitation downscaling improves the coarse resolution and poor representation of precipitation in global climate models and helps end users to assess the likely hydrological impacts of climate change. This paper integrates perspectives from meteorologists, climatologists, statisticians, and hydrologists to identify generic end user (in particular, impact modeler) needs and to discuss downscaling capabilities and gaps. End users need a reliable representation of precipitation intensities and temporal and spatial variability, as well as physical consistency, independent of region and season. In addition to presenting dynamical downscaling, we review perfect prognosis statistical downscaling, model output statistics, and weather generators, focusing on recent developments to improve the representation of space-time variability. Furthermore, evaluation techniques to assess downscaling skill are presented. Downscaling adds considerable value to projections from global climate models. Remaining gaps are uncertainties arising from sparse data; representation of extreme summer precipitation, subdaily precipitation, and full precipitation fields on fine scales; capturing changes in small-scale processes and their feedback on large scales; and errors inherited from the driving global climate model.
Abstract The role of personality traits in the decision to start a business and to maintain it successfully is discussed controversially in entrepreneurship research. Our meta-analysis builds upon and extends earlier meta-analyses by doing a full analysis of personality traits that includes a comparison of different traits from a theoretical perspective and by analysing a full set of personality predictors for both start-up activities as well as success. Theoretically, our article adds to the literature by matching traits to the tasks of entrepreneurs. The results indicate that traits matched to the task of running a business produced higher effect sizes with business creation than traits that were not matched to the task of running an enterprise, corrected r = .247, K = 47, N = 13,280, and corrected r = .124, K = 20, N = 3975, respectively. Moreover, traits matched to the task produced higher correlations with success, corrected r = .250, K = 42, N = 5607, than traits not matched to the task of running a business, corrected r = .028, K = 13, N = 2777. The traits matched to entrepreneurship significantly correlated with entrepreneurial behaviour (business creation, business success) were need for achievement, generalized self-efficacy, innovativeness, stress tolerance, need for autonomy, and proactive personality. These relationships were of moderate size in general and, moreover, heterogeneity suggested that future research should analyse moderator variables. Abstract This paper contributes to the discussion on contingencies of process innovations by focusing on and introducing organizational‐level constructs of climate for initiative and psychological safety. We argue that process innovations, defined as deliberate and new organizational attempts to change production and service processes, need to be accompanied by climates that complement the adoption and implementation of such innovations. Our study of 47 mid‐sized German companies examines the relation between process innovations, climates for initiative and psychological safety, and firm performance. Results show that climates for initiative and psychological safety were positively related to two measures of firm performance—longitudinal change in return on assets (holding prior return on assets constant) and firm goal achievement—and moderated the relation between process innovations and firm performance. Tribolium castaneum is a member of the most species-rich eukaryotic order, a powerful model organism for the study of generalized insect development, and an important pest of stored agricultural products. We describe its genome sequence here. This omnivorous beetle has evolved the ability to interact with a diverse chemical environment, as shown by large expansions in odorant and gustatory receptors, as well as P450 and other detoxification enzymes. Development in Tribolium is more representative of other insects than is Drosophila, a fact reflected in gene content and function. For example, Tribolium has retained more ancestral genes involved in cell–cell communication than Drosophila, some being expressed in the growth zone crucial for axial elongation in short-germ development. Systemic RNA interference in T. castaneum functions differently from that in Caenorhabditis elegans, but nevertheless offers similar power for the elucidation of gene function and identification of targets for selective insect control. The red flour beetle Tribolium castaneum is a common pest: a type of 'bran bug', it targets cereal products, including grain, flour and rice bran. It is also a commonly used laboratory model, combining the ease of systematic RNA interference experiments such as those used with the nematode worm C. elegans with a biology that is more representative of most insects than even Drosophila. This weeks sees the publication by the Tribolium Genome Sequencing Consortium of the genomic sequence of T. castaneum. This is the first beetle genome to be published, and it will be a valuable resource for insect development studies and pest biology. The beetle Tribolium castaneum is a commonly used laboratory model, combining the ease of systematic RNAi experiments like those in Caenorhabditis elegans, with biology that is more representative of most insects than Drosophila melanogaster. A large consortium has sequenced and analysed the genome of the red flour beetle, creating a resource for biologists everywhere.

Periodontal health is defined by absence of clinically detectable inflammation. There is a biological level of immune surveillance that is consistent with clinical gingival health and homeostasis. Clinical gingival health may be found in a periodontium that is intact, i.e. without clinical attachment loss or bone loss, and on a reduced periodontium in either a non-periodontitis patient (e.g. in patients with some form of gingival recession or following crown lengthening surgery) or in a patient with a history of periodontitis who is currently periodontally stable. Clinical gingival health can be restored following treatment of gingivitis and periodontitis. However, the treated and stable periodontitis patient with current gingival health remains at increased risk of recurrent periodontitis, and accordingly, must be closely monitored. Two broad categories of gingival diseases include non-dental plaque biofilm-induced gingival diseases and dental plaque-induced gingivitis. Non-dental plaque biofilm-induced gingival diseases include a variety of conditions that are not caused by plaque and usually do not resolve following plaque removal. Such lesions may be manifestations of a systemic condition or may be localized to the oral cavity. Dental plaque-induced gingivitis has a variety of clinical signs and symptoms, and both local predisposing factors and systemic modifying factors can affect its extent, severity, and progression. Dental plaque-induced gingivitis may arise on an intact periodontium or on a reduced periodontium in either a non-periodontitis patient or in a currently stable "periodontitis patient" i.e. successfully treated, in whom clinical inflammation has been eliminated (or substantially reduced). A periodontitis patient with gingival inflammation remains a periodontitis patient (Figure 1), and comprehensive risk assessment and management are imperative to ensure early prevention and/or treatment of recurrent/progressive periodontitis. Precision dental medicine defines a patient-centered approach to care, and therefore, creates differences in the way in which a "case" of gingival health or gingivitis is defined for clinical practice as opposed to epidemiologically in population prevalence surveys. Thus, case definitions of gingival health and gingivitis are presented for both purposes. While gingival health and gingivitis have many clinical features, case definitions are primarily predicated on presence or absence of bleeding on probing. Here we classify gingival health and gingival diseases/conditions, along with a summary table of diagnostic features for defining health and gingivitis in various clinical situations.
The predictive power of the Ajzen, Triandis, and Schwartz models are compared in the context of car use for university routes. Two hundred fifty-four students filled out a questionnaire designed to measure the components of the three models. In the prediction of intention to use a car, results indicated that one variable from the Trinandis model—role beliefs—increased the explanatory power offered by the components of the Ajzen model. In the prediction of self-reported car use, one variable of the Triandis model—car use habit—significantly increased the predictive power of the Ajzen model. The central variable of the Schwartz model—personal norm—exerted no significant effect either on intention or on behavior. The implications of the findings for interventions to reduce the car use of students for university routes are discussed.

The authors argue that a high-organizational error management culture, conceptualized to include norms and common practices in organizations (e.g., communicating about errors, detecting, analyzing, and correcting errors quickly), is pivotal to the reduction of negative and the promotion of positive error consequences. Organizational error management culture was positively related to firm performance across 2 studies conducted in 2 different European countries. On the basis of quantitative and qualitative cross-sectional data from 65 Dutch organizations, Study 1 revealed that organizational error management culture was significantly correlated with both organizational goal achievement and an objective indicator of economic performance. This finding was confirmed in Study 2, using change-of-profitability data from 47 German organizations. The results suggest that organizations may want to introduce organizational error management as a way to boost firm performance.
BACKGROUND: Current methods used to restore the joint surface in patients with localized articular cartilage defects include transplantation of an autologous osteochondral cylinder and implantation of autologous chondrocytes. The purpose of this study was to evaluate the clinical and histological outcomes of these two techniques. METHODS: We performed a prospective clinical study to investigate the two-year outcomes in forty patients with an articular cartilage lesion of the femoral condyle who had been randomly treated with either transplantation of an autologous osteochondral cylinder or implantation of autologous chondrocytes. Biopsy specimens from representative patients of both groups were evaluated with histological staining, immunohistochemistry, and scanning electron microscopy. RESULTS: According to the postoperative Lysholm score, the recovery after autologous chondrocyte implantation was slower than that after osteochondral transplantation at six months (p < or = 0.015), twelve months (p < or = 0.001), and twenty-four months (p < or = 0.012). On the basis of the Meyers score and the Tegner activity score, the results were equally good with the two methods two years after treatment. Histomorphological evaluation of biopsy specimens within two years after autologous chondrocyte implantation demonstrated a complete, mechanically stable resurfacing of the defect in all patients. The tissue consisted mainly of fibrocartilage, while localized areas of hyaline-like regenerative cartilage could be detected close to the subchondral bone. Although a gap remained at the site of the transplantation in all five biopsy specimens examined as long as two years after osteochondral cylinder transplantation, histomorphological analysis and scanning electron microscopy revealed no differences between the osteochondral transplants and the surrounding original cartilage. CONCLUSIONS: Both treatments resulted in a decrease in symptoms. However, the improvement provided by the autologous chondrocyte implantation lagged behind that provided by the osteochondral cylinder transplantation. Histologically, the defects treated with autologous chondrocyte implantation were primarily filled with fibrocartilage, whereas the osteochondral cylinder transplants retained their hyaline character, although there was a persistent interface between the transplant and the surrounding original cartilage. Limitations of our study included the small number of patients, the relatively short (two-year) follow-up, and the absence of a control group.
Abstract This article analyses the relationship between mobbing, job characteristics, social environment variables, and psychological ill-health. The Leymann Inventory of Psychological Terrorization (LIFT) was factor analysed and led to seven factors in two samples of mobbing victims (N = 50 and N = 99): Mobbing by organizational measures, social isolation, attacking the victim's private life, attacking the victim's attitudes, physical violence, verbal aggression, and rumours. Mobbing was correlated with bad job content, a bad social environment, and psychological ill-health. The findings suggest that the more social support supervisors gave, the less the victims reported being shouted at, being constantly criticized, and receiving verbal threats. In contrast, the more social support the victims received from their colleagues the less they reported being socially isolated or being ridiculed with regard to their private life. Moreover, having private life attacked showed the strongest correlation with psychological ill-health. The data suggest that organizational factors are potential causes of mobbing at work.
The Schwartz (1992) theory of basic human values has promoted a revival of empirical research on values. The semi-annual European Social Survey (ESS) includes a new 21-item instrument to measure the importance of the 10 basic values of the theory. Representative national samples in 20 countries responded to the instrument in 2002–3. We briefly describe the theory and the ESS instrument and assess its adequacy for measuring values across countries. Using multiple-group confirmatory factor analyses, augmented with mean-structure information, we assess the configural and measurement (metric) invariance of the values—necessary conditions for equivalence of the meaning of constructs and scalar invariance—a precondition for comparing value means across countries. Only if such equivalence is established can researchers make meaningful and clearly interpretable cross-national comparisons of value priorities and their correlates. The ESS values scale demonstrates configural and metric invariance, allowing researchers to use it to study relationships among values, attitudes, behavior and socio-demographic characteristics across countries. Comparing the mean importance of values across countries is possible only for subsets of countries where scalar invariance holds.
Autoimmunity is complicated by bone loss. In human rheumatoid arthritis (RA), the most severe inflammatory joint disease, autoantibodies against citrullinated proteins are among the strongest risk factors for bone destruction. We therefore hypothesized that these autoantibodies directly influence bone metabolism. Here, we found a strong and specific association between autoantibodies against citrullinated proteins and serum markers for osteoclast-mediated bone resorption in RA patients. Moreover, human osteoclasts expressed enzymes eliciting protein citrullination, and specific N-terminal citrullination of vimentin was induced during osteoclast differentiation. Affinity-purified human autoantibodies against mutated citrullinated vimentin (MCV) not only bound to osteoclast surfaces, but also led to robust induction of osteoclastogenesis and bone-resorptive activity. Adoptive transfer of purified human MCV autoantibodies into mice induced osteopenia and increased osteoclastogenesis. This effect was based on the inducible release of TNF-α from osteoclast precursors and the subsequent increase of osteoclast precursor cell numbers with enhanced expression of activation and growth factor receptors. Our data thus suggest that autoantibody formation in response to citrullinated vimentin directly induces bone loss, providing a link between the adaptive immune system and bone.
We present a comprehensive review of methods to measure soil water content with ground penetrating radar (GPR). We distinguish four methodologies: soil water content determined from reflected wave velocity, soil water content determined from ground wave velocity, soil water content determined from transmitted wave velocity between boreholes, and soil water content determined from the surface reflection coefficient. For each of these four methodologies, we discuss the basic principles, illustrate the quality of the data with field examples, discuss the possibilities and limitations, and identify areas where future research is required. We hope that this review will further stimulate the community to consider ground penetrating radar as one of the possible tools to measure soil water content.

Numerous studies showed decreased performance in situations that require multiple tasks or actions relative to appropriate control conditions. Because humans often engage in such multitasking activities, it is important to understand how multitasking affects performance. In the present article, we argue that research on dual-task interference and sequential task switching has proceeded largely separately using different experimental paradigms and methodology. In our article we aim at organizing this complex set of research in terms of three complementary research perspectives on human multitasking. One perspective refers to structural accounts in terms of cognitive bottlenecks (i.e., critical processing stages). A second perspective refers to cognitive flexibility in terms of the underlying cognitive control processes. A third perspective emphasizes cognitive plasticity in terms of the influence of practice on human multitasking abilities. With our review article we aimed at highlighting the value of an integrative position that goes beyond isolated consideration of a single theoretical research perspective and that broadens the focus from single experimental paradigms (dual task and task switching) to favor instead a view that emphasizes the fundamental similarity of the underlying cognitive mechanisms across multitasking paradigms.
