We developed a set of universal PCR primers (MiFish-U/E) for metabarcoding environmental DNA (eDNA) from fishes. Primers were designed using aligned whole mitochondrial genome (mitogenome) sequences from 880 species, supplemented by partial mitogenome sequences from 160 elasmobranchs (sharks and rays). The primers target a hypervariable region of the 12S rRNA gene (163-185 bp), which contains sufficient information to identify fishes to taxonomic family, genus and species except for some closely related congeners. To test versatility of the primers across a diverse range of fishes, we sampled eDNA from four tanks in the Okinawa Churaumi Aquarium with known species compositions, prepared dual-indexed libraries and performed paired-end sequencing of the region using high-throughput next-generation sequencing technologies. Out of the 180 marine fish species contained in the four tanks with reference sequences in a custom database, we detected 168 species (93.3%) distributed across 59 families and 123 genera. These fishes are not only taxonomically diverse, ranging from sharks and rays to higher teleosts, but are also greatly varied in their ecology, including both pelagic and benthic species living in shallow coastal to deep waters. We also sampled natural seawaters around coral reefs near the aquarium and detected 93 fish species using this approach. Of the 93 species, 64 were not detected in the four aquarium tanks, rendering the total number of species detected to 232 (from 70 families and 152 genera). The metabarcoding approach presented here is non-invasive, more efficient, more cost-effective and more sensitive than the traditional survey methods. It has the potential to serve as an alternative (or complementary) tool for biodiversity monitoring that revolutionizes natural resource management and ecological studies of fish communities on larger spatial and temporal scales.
Environmental DNA (eDNA) from aquatic vertebrates has recently been used to estimate the presence of a species. We hypothesized that fish release DNA into the water at a rate commensurate with their biomass. Thus, the concentration of eDNA of a target species may be used to estimate the species biomass. We developed an eDNA method to estimate the biomass of common carp (Cyprinus carpio L.) using laboratory and field experiments. In the aquarium, the concentration of eDNA changed initially, but reached an equilibrium after 6 days. Temperature had no effect on eDNA concentrations in aquaria. The concentration of eDNA was positively correlated with carp biomass in both aquaria and experimental ponds. We used this method to estimate the biomass and distribution of carp in a natural freshwater lagoon. We demonstrated that the distribution of carp eDNA concentration was explained by water temperature. Our results suggest that biomass data estimated from eDNA concentration reflects the potential distribution of common carp in the natural environment. Measuring eDNA concentration offers a non-invasive, simple, and rapid method for estimating biomass. This method could inform management plans for the conservation of ecosystems.
We report on the population properties of compact binary mergers inferred from gravitational-wave observations of these systems during the first three LIGO-Virgo observing runs. The Gravitational-Wave Transient Catalog 3 (GWTC-3) contains signals consistent with three classes of binary mergers: binary black hole, binary neutron star, and neutron star–black hole mergers. We infer the binary neutron star merger rate to be between 10 and <a:math xmlns:a="http://www.w3.org/1998/Math/MathML" display="inline"><a:mrow><a:mn>1700</a:mn><a:mtext> </a:mtext><a:mtext> </a:mtext><a:msup><a:mrow><a:mi>Gpc</a:mi></a:mrow><a:mrow><a:mo>−</a:mo><a:mn>3</a:mn></a:mrow></a:msup><a:mtext> </a:mtext><a:msup><a:mrow><a:mi>yr</a:mi></a:mrow><a:mrow><a:mo>−</a:mo><a:mn>1</a:mn></a:mrow></a:msup></a:mrow></a:math> and the neutron star–black hole merger rate to be between 7.8 and <c:math xmlns:c="http://www.w3.org/1998/Math/MathML" display="inline"><c:mrow><c:mn>140</c:mn><c:mtext> </c:mtext><c:mtext> </c:mtext><c:msup><c:mrow><c:mi>Gpc</c:mi></c:mrow><c:mrow><c:mo>−</c:mo><c:mn>3</c:mn></c:mrow></c:msup><c:mtext> </c:mtext><c:msup><c:mrow><c:mi>yr</c:mi></c:mrow><c:mrow><c:mo>−</c:mo><c:mn>1</c:mn></c:mrow></c:msup></c:mrow></c:math>, assuming a constant rate density in the comoving frame and taking the union of 90% credible intervals for methods used in this work. Using both binary neutron star and neutron star–black hole binaries, we obtain a broad, relatively flat neutron star mass distribution extending from <q:math xmlns:q="http://www.w3.org/1998/Math/MathML" display="inline"><q:msubsup><q:mn>1.2</q:mn><q:mrow><q:mo>−</q:mo><q:mn>0.2</q:mn></q:mrow><q:mrow><q:mo>+</q:mo><q:mn>0.1</q:mn></q:mrow></q:msubsup></q:math> to <s:math xmlns:s="http://www.w3.org/1998/Math/MathML" display="inline"><s:msubsup><s:mn>2.0</s:mn><s:mrow><s:mo>−</s:mo><s:mn>0.3</s:mn></s:mrow><s:mrow><s:mo>+</s:mo><s:mn>0.3</s:mn></s:mrow></s:msubsup><s:msub><s:mi>M</s:mi><s:mo stretchy="false">⊙</s:mo></s:msub></s:math>. We confidently determine that the merger rate as a function of mass sharply declines after the expected maximum neutron star mass, but cannot yet confirm or rule out the existence of a lower mass gap between neutron stars and black holes. We also find the binary black hole mass distribution has localized over- and underdensities relative to a power-law distribution, with peaks emerging at chirp masses of <v:math xmlns:v="http://www.w3.org/1998/Math/MathML" display="inline"><v:msubsup><v:mn>8.3</v:mn><v:mrow><v:mo>−</v:mo><v:mn>0.5</v:mn></v:mrow><v:mrow><v:mo>+</v:mo><v:mn>0.3</v:mn></v:mrow></v:msubsup></v:math> and <x:math xmlns:x="http://www.w3.org/1998/Math/MathML" display="inline"><x:msubsup><x:mn>27.9</x:mn><x:mrow><x:mo>−</x:mo><x:mn>1.8</x:mn></x:mrow><x:mrow><x:mo>+</x:mo><x:mn>1.9</x:mn></x:mrow></x:msubsup><x:msub><x:mi>M</x:mi><x:mo stretchy="false">⊙</x:mo></x:msub></x:math>. While we continue to find that the mass distribution of a binary’s more massive component strongly decreases as a function of primary mass, we observe no evidence of a strongly suppressed merger rate above approximately <ab:math xmlns:ab="http://www.w3.org/1998/Math/MathML" display="inline"><ab:mn>60</ab:mn><ab:msub><ab:mi>M</ab:mi><ab:mo stretchy="false">⊙</ab:mo></ab:msub></ab:math>, which would indicate the presence of a upper mass gap. Observed black hole spins are small, with half of spin magnitudes below <db:math xmlns:db="http://www.w3.org/1998/Math/MathML" display="inline"><db:msub><db:mi>χ</db:mi><db:mi>i</db:mi></db:msub><db:mo>≈</db:mo><db:mn>0.25</db:mn></db:math>. While the majority of spins are preferentially aligned with the orbital angular momentum, we infer evidence of antialigned spins among the binary population. We observe an increase in spin magnitude for systems with more unequal-mass ratio. We also observe evidence of misalignment of spins relative to the orbital angular momentum. Ecological theory predicts that a complex community formed by a number of species is inherently unstable, guiding ecologists to identify what maintains species diversity in nature. Earlier studies often assumed a community with only one interaction type, either an antagonistic, competitive, or mutualistic interaction, leaving open the question of what the diversity of interaction types contributes to the community maintenance. We show theoretically that the multiple interaction types might hold the key to understanding community dynamics. A moderate mixture of antagonistic and mutualistic interactions can stabilize population dynamics. Furthermore, increasing complexity leads to increased stability in a "hybrid" community. We hypothesize that the diversity of species and interaction types may be the essential element of biodiversity that maintains ecological communities.

We report the observation of gravitational waves from two compact binary coalescences in LIGO's and Virgo's third observing run with properties consistent with neutron star-black hole (NSBH) binaries. The two events are named GW200105_162426 and GW200115_042309, abbreviated as GW200105 and GW200115; the first was observed by LIGO Livingston and Virgo and the second by all three LIGO-Virgo detectors. The source of GW200105 has component masses, whereas the source of GW200115 has component masses and (all measurements quoted at the 90% credible level). The probability that the secondary's mass is below the maximal mass of a neutron star is 89%-96% and 87%-98%, respectively, for GW200105 and GW200115, with the ranges arising from different astrophysical assumptions. The source luminosity distances are and, respectively. The magnitude of the primary spin of GW200105 is less than 0.23 at the 90% credible level, and its orientation is unconstrained. For GW200115, the primary spin has a negative spin projection onto the orbital angular momentum at 88% probability. We are unable to constrain the spin or tidal deformation of the secondary component for either event. We infer an NSBH merger rate density of when assuming that GW200105 and GW200115 are representative of the NSBH population or under the assumption of a broader distribution of component masses. © 2021. The phenomenal increase during the past decade in research utilizing pulsed electric current to activate sintering is attributed generally to the intrinsic advantages of the method relative to conventional sintering methods and to the observations of the enhanced properties of materials consolidated by this method. This review focuses on the fundamental aspects of the process, discussing the reported observations and simulation studies in terms of the basic aspects of the process and identifying the intrinsic benefits of the use of the parameters of current (and pulsing), pressure, and heating rate.
Plants are constantly exposed to a variety of environmental stresses. Freezing or extremely low temperature constitutes a key factor influencing plant growth, development and crop productivity. Plants have evolved a mechanism to enhance tolerance to freezing during exposure to periods of low, but non-freezing temperatures. This phenomenon is called cold acclimation. During cold acclimation, plants develop several mechanisms to minimize potential damages caused by low temperature. Cold response is highly complex process that involves an array of physiological and biochemical modifications. Furthermore, alterations of the expression patterns of many genes, proteins and metabolites in response to cold stress have been reported. Recent studies demonstrate that post-transcriptional and post-translational regulations play a role in the regulation of cold signaling. In this review article, recent advances in cold stress signaling and tolerance are highlighted.
Abstract Environmental DNA (eDNA) metabarcoding has emerged as a potentially powerful tool to assess aquatic community structures. However, the method has hitherto lacked field tests that evaluate its effectiveness and practical properties as a biodiversity monitoring tool. Here, we evaluated the ability of eDNA metabarcoding to reveal fish community structures in species-rich coastal waters. High-performance fish-universal primers and systematic spatial water sampling at 47 stations covering ~11 km 2 revealed the fish community structure at a species resolution. The eDNA metabarcoding based on a 6-h collection of water samples detected 128 fish species, of which 62.5% (40 species) were also observed by underwater visual censuses conducted over a 14-year period. This method also detected other local fishes (≥23 species) that were not observed by the visual censuses. These eDNA metabarcoding features will enhance marine ecosystem-related research, and the method will potentially become a standard tool for surveying fish communities.
The recent detections of gravitational waves (GWs) reported by LIGO/Virgo collaborations have made significant impact on physics and astronomy. A global network of GW detectors will play a key role to solve the unknown nature of the sources in coordinated observations with astronomical telescopes and detectors. Here we introduce KAGRA (former name LCGT; Large-scale Cryogenic Gravitational wave Telescope), a new GW detector with two 3-km baseline arms arranged in the shape of an "L", located inside the Mt. Ikenoyama, Kamioka, Gifu, Japan. KAGRA's design is similar to those of the second generations such as Advanced LIGO/Virgo, but it will be operating at the cryogenic temperature with sapphire mirrors. This low temperature feature is advantageous for improving the sensitivity around 100 Hz and is considered as an important feature for the third generation GW detector concept (e.g. Einstein Telescope of Europe or Cosmic Explorer of USA). Hence, KAGRA is often called as a 2.5 generation GW detector based on laser interferometry. The installation and commissioning of KAGRA is underway and its cryogenic systems have been successfully tested in May, 2018. KAGRA's first observation run is scheduled in late 2019, aiming to join the third observation run (O3) of the advanced LIGO/Virgo network. In this work, we describe a brief history of KAGRA and highlights of main feature. We also discuss the prospects of GW observation with KAGRA in the era of O3. When operating along with the existing GW detectors, KAGRA will be helpful to locate a GW source more accurately and to determine the source parameters with higher precision, providing information for follow-up observations of a GW trigger candidate.
A great deal of research has been performed recently on robots that feature functions for communicating with humans in daily life, i.e., communication robots. We consider it important to develop methods to measure humans’ attitudes and emotions that may prevent them from interaction with communication robots, as indices to study short-term and long-term interaction between humans and communication robots. This study is aimed at exploring the influence of negative attitudes toward robots, focusing on applications of communication robots to daily-life services. First, a scale of negative attitudes toward robots consisting of three subordinate scales, “negative attitudes toward situations of interaction with robots,” “negative attitudes toward the social influence of robots,” and “negative attitudes toward emotions in interaction with robots,” was developed based on a data sample comprising of 263 Japanese university students. This scale was administered to 240 Japanese university students to confirm its validity and reliability. In this paper, we report on the results of analyses of these data samples. Moreover, we discuss some future problems including a comparison of attitudes toward robots between nations.
We show a direct connection between a cellular automaton and integrable nonlinear wave equations. We also present the $N$-soliton formula for the cellular automaton. Finally, we propose a general method for constructing such integrable cellular automata and their $N$-soliton solutions.
Turning noise into useful data—every geophysicist's dream? And now it seems possible. The field of seismic interferometry has at its foundation a shift in the way we think about the parts of the signal that are currently filtered out of most analyses—complicated seismic codas (the multiply scattered parts of seismic waveforms) and background noise (whatever is recorded when no identifiable active source is emitting, and which is superimposed on all recorded data). Those parts of seismograms consist of waves that reflect and refract around exactly the same subsurface heterogeneities as waves excited by active sources. The key to the rapid emergence of this field of research is our new understanding of how to unravel that subsurface information from these relatively complex-looking waveforms. And the answer turned out to be rather simple. This article explains the operation of seismic interferometry and provides a few examples of its application.
We report results of a search for an isotropic gravitational-wave background (GWB) using data from Advanced LIGO's and Advanced Virgo's third observing run (O3) combined with upper limits from the earlier O1 and O2 runs. Unlike in previous observing runs in the advanced detector era, we include Virgo in the search for the GWB. The results of the search are consistent with uncorrelated noise, and therefore we place upper limits on the strength of the GWB. We find that the dimensionless energy density GW 5.8 10 -9 at the 95% credible level for a flat (frequency-independent) GWB, using a prior which is uniform in the log of the strength of the GWB, with 99% of the sensitivity coming from the band 20-76.6 Hz; GW f 3.4 10 -9 at 25 Hz for a power-law GWB with a spectral index of 2=3 (consistent with expectations for compact binary coalescences), in the band 20-90.6 Hz; and GW f 3.9 10 -10 at 25 Hz for a spectral index of 3, in the band 20-291.6 Hz. These upper limits improve over our previous results by a factor of 6.0 for a flat GWB, 8.8 for a spectral index of 2=3, and 13.1 for a spectral index of 3. We also search for a GWB arising from scalar and vector modes, which are predicted by alternative theories of gravity; we do not find evidence of these, and place upper limits on the strength of GWBs with these polarizations. We demonstrate that there is no evidence of correlated noise of magnetic origin by performing a Bayesian analysis that allows for the presence of both a GWB and an effective magnetic background arising from geophysical Schumann resonances. We compare our upper limits to a fiducial model for the GWB from the merger of compact binaries, updating the model to use the most recent datadriven population inference from the systems detected during O3a. Finally, we combine our results with observations of individual mergers and show that, at design sensitivity, this joint approach may yield stronger constraints on the merger rate of binary black holes at z 2 than can be achieved with individually resolved mergers alone.
Abstract The Deci-hertz Interferometer Gravitational Wave Observatory (DECIGO) is a future Japanese space mission with a frequency band of 0.1 Hz to 10 Hz. DECIGO aims at the detection of primordial gravitational waves, which could have been produced during the inflationary period right after the birth of the Universe. There are many other scientific objectives of DECIGO, including the direct measurement of the acceleration of the expansion of the Universe, and reliable and accurate predictions of the timing and locations of neutron star/black hole binary coalescences. DECIGO consists of four clusters of observatories placed in heliocentric orbit. Each cluster consists of three spacecraft, which form three Fabry–Pérot Michelson interferometers with an arm length of 1000 km. Three DECIGO clusters will be placed far from each other, and the fourth will be placed in the same position as one of the other three to obtain correlation signals for the detection of primordial gravitational waves. We plan to launch B-DECIGO, which is a scientific pathfinder for DECIGO, before DECIGO in the 2030s to demonstrate the technologies required for DECIGO, as well as to obtain fruitful scientific results to further expand multi-messenger astronomy.
When people interact with communication robots in daily life, their attitudes and emotions toward the robots affect their behavior. From the perspective of robotics design, we need to investigate the influences of these attitudes and emotions on human-robot interaction. This paper reports our empirical study on the relationships between people's attitudes and emotions, and their behavior toward a robot. In particular, we focused on negative attitudes, anxiety, and communication avoidance behavior, which have important implications for robotics design. For this purpose, we used two psychological scales that we had developed: negative attitudes toward robots scale (NARS) and robot anxiety scale (RAS). In the experiment, subjects and a humanoid robot are engaged in simple interactions including scenes of meeting, greeting, self-disclosure, and physical contact. Experimental results indicated that there is a relationship between negative attitudes and emotions, and communication avoidance behavior. A gender effect was also suggested.
The environmental DNA (eDNA) technique is expected to become a powerful, non-invasive tool for estimating the distribution and biomass of organisms. This technique was recently shown to be applicable to aquatic vertebrates by collecting extraorganismal DNA floating in the water or absorbed onto suspended particles. However, basic information on eDNA release rate is lacking, despite it being essential for practical applications. In this series of experiments with bluegill sunfish (Lepomis macrochirus), we examined the effect of fish developmental stage on eDNA release rate. eDNA concentration reached equilibrium 3 days after the individual fish were introduced into the separate containers, enabling calculation of the eDNA release rate (copies h-1) from individual fish on the assumption that the number of eDNA released from the fish per unit time equals total degradation in the container (copies h-1). The eDNA release rate was 3-4 times higher in the adult (body weight: 30-75 g) than in the juvenile group (0.5-2.0 g). Such positive relationship between fish size and eDNA release rate support the possibility of biomass rather than density estimation using eDNA techniques. However, the eDNA release rate per fish body weight (copies h-1 g-1) was slightly higher in the juvenile than the adult group, which is likely because of the ontogenetic reduction in metabolic activity. Therefore, quantitative eDNA data should be carefully interpreted to avoid overestimating biomass when the population is dominated by juveniles, because the age structure of the focal population is often variable and unseen in the field. eDNA degradation rates (copies l-1 h-1), calculated by curve fitting of time-dependent changes in eDNA concentrations after fish removal, were 5.1-15.9% per hour (half-life: 6.3 h). This suggests that quantitative eDNA data should be corrected using a degradation curve attained in the target field.
There are various recommendations by many coaches regarding foot landing techniques in distance running that are meant to improve running performance and prevent injuries. Several studies have investigated the kinematic and kinetic differences between rearfoot strike (RFS), midfoot strike (MFS), and forefoot strike (FFS) patterns at foot landing and their effects on running efficiency on a treadmill and over ground conditions. However, little is known about the actual condition of the foot strike pattern during an actual road race at the elite level of competition. The purpose of the present study was to document actual foot strike patterns during a half marathon in which elite international level runners, including Olympians, compete. Four hundred fifteen runners were filmed by 2 120-Hz video cameras in the height of 0.15 m placed at the 15.0-km point and obtained sagittal foot landing and taking off images for 283 runners. Rearfoot strike was observed in 74.9% of all analyzed runners, MFS in 23.7%, and FFS in 1.4%. The percentage of MFS was higher in the faster runners group, when all runners were ranked and divided into 50 runner groups at the 15.0-km point of the competition. In the top 50, which included up to the 69th place runner in actual order who passed the 15-km point at 45 minutes, 53 second (this speed represents 5.45 m x s(-1), or 15 minutes, 17 seconds per 5 km), RFS, MFS, and FFS were 62.0, 36.0, and 2.0%, respectively. Contact time (CT) clearly increased for the slower runners, or the placement order increased (r = 0.71, p < or = 0.05). The CT for RFS + FFS for every 50 runners group significantly increased with increase of the placement order. The CT for RFS was significantly longer than MFS + FFS (200.0 +/- 21.3 vs. 183.0 +/- 16 millisecond). Apparent inversion (INV) of the foot at the foot strike was observed in 42% of all runners. The percentage of INV for MFS was higher than for RFS and FFS (62.5, 32.0, and 50%, respectively). The CT with INV for MFS + FFS was significantly shorter than the CT with and without INV for RFS. Furthermore, the CT with INV was significantly shorter than push-off time without INV for RFS. The findings of this study indicate that foot strike patterns are related to running speed. The percentage of RFS increases with the decreasing of the running speed; conversely, the percentage of MFS increases as the running speed increases. A shorter contact time and a higher frequency of inversion at the foot contact might contribute to higher running economy.

Summary Environmental DNA ( eDNA ) analysis for detecting the presence of aquatic and terrestrial organisms is an established method, and the eDNA concentration of a species can reflect its abundance/biomass at a site. However, attempts to estimate the abundance/biomass of aquatic species using eDNA concentrations in large stream and river ecosystems have received little attention. We determined the eDNA concentration and abundance/biomass of a stream fish, Plecoglossus altivelis , by conducting a snorkelling survey in the Saba River, Japan. Furthermore, we evaluated the relationship between eDNA concentrations and the estimated abundance/biomass of P. altivelis , and determined its spatial distribution within the river. Across the three seasons from spring to autumn, we found significant correlations between the eDNA concentration of P. altivelis and its abundance/biomass at study sites within the river. We detected the eDNA at the sites where we found only feeding traces on stones (where P. altivelis was not directly observed), but not at sites without feeding traces. Additionally, we tested the optimal number of qPCR replicates needed for the eDNA evaluation of P. altivelis abundance and biomass; only a small number of replicates was required when the eDNA concentration was high. Our findings suggest that eDNA analysis is a useful tool to estimate fish abundance/biomass as well as their spatial distribution in rivers.
